DSHB Data Sheet

Catalog Fields

Antigen: Fasciclin III (Drosophila)
Hybridoma Cells Available: Yes
Antigen Species: Drosophila
Depositor: Goodman, C.
Isotype: MIgG2a
Depositors Institution: University of California, Berkeley
Host Species: mouse
Depositors Notes: Fas III is expressed on a subset of neurons and axons in the CNS, patches of ectoderm, ectodermal stripes, parts of the hindgut and foregut, visceral mesoderm, and the salivary gland. The antibody can be used to stain dissections without detergent as the epitope is on the extracellular domain of the protein. After 10 hrs in development, ectodermal staining will obscure CNS in whole mount preps and therefore dissected preparations are recommended for embryos staining past this age. During larval development, staining is present on a subset of neurons, the epithelial layer of the discs and a cyclone fence pattern in the eye disc. It is especially useful for following the development of the RP1 and RP3 neurons. The antibody works for affinity purification but does not work for immunoprecipitation or western.
Positive Tested Species Reactivity: Drosophila
Gene: Fas3
Antigen Molecular Weight: Multiple isoforms; Apparent: 46, 59, 66, and 80 kDa
Alternate Gene Names: Fasciclin 3
Immunogen: Membrane preparation from nerve cord and cultures of primary neurons (10-13 hr development).
Clonality: Monoclonal
Myeloma Strain: NS-1
Epitope Mapped: No
Uniprot ID: P15278
Additional Information: This antibody may work for Immunoprecipitation. While the Depositor clearly stated that it doesn't work for immunoprecipitation (see Depositor Notes), the Initial Publication, in which the Depositor is the last author, reports the succesful use of 7G10 for the immunoprecipitation of Fasciclin III.
Entrez Gene ID: 35097
Antibody Registry ID: AB_528238
Recommended Applications: Immunofluorescence, Immunohistochemistry, Immunoprecipitation

Storage and Handling Recommendations
Although many cell products are maintained at 4°C for years without loss of activity, shelf-life at 4°C is highly variable. To ensure retention of antibody activity, we recommend aliquotting the product into two parts: 1) a volume of antibody stored at 4°C to be used within two weeks. 2) the remaining product diluted with an equal volume of molecular grade glycerol and stored at -20°C.
Usage Recommendations
While optimal Ig concentration for an application will vary, a good starting concentration for immunohistochemistry (IHC), immunofluorescence(IF) and staining is 2-5 µg/ml. For Western blots, the concentration is decreased by one order of magnitude (that is, 0.2-0.5 µg/ml).
